DESTINY-Lung04
First-line (1L) trastuzumab deruxtecan (T-DXd) in patients (pts) with HER2-mutant (HER2m) NSCLC: Patient-reported outcomes (PROs) from DESTINY-Lung04 (DL-04)
In patients with untreated advanced HER2-mutant NSCLC, trastuzumab deruxtecan delayed deterioration in patient-reported cough compared with pembrolizumab plus chemotherapy.
Patient-reported cough deteriorated later with trastuzumab deruxtecan, but the HR's upper confidence limit of 0.99 only just excludes no effect, and the abstract gives no P values or multiplicity control for these secondary endpoints. The dyspnoea and chest pain HRs have confidence intervals that cross 1. The trastuzumab deruxtecan chest pain median and several upper confidence limits are not estimable. Because the trial was open label, patients knew their treatment, which can bias self-reported symptoms and tolerability. Median treatment duration was much longer with trastuzumab deruxtecan, so on-treatment comparisons from Week 20 and at end of treatment may reflect who remained on therapy, and symptomatic AE data are deferred to the presentation.
